چکیده
Abstract Complaints of control failures with acetolactate synthase (ALS)- and protoporphyrinogen oxidase (PPO)-inhibiting herbicides on redroot pigweed ( Amaranthus retroflexus L.) were reported in conventional soybean [ Glycine max (L.) Merr.] fields North Carolina. Greenhouse dose–response assays confirmed that the Camden County Pasquotank populations less sensitive to ALS- PPO-inhibiting compared susceptible A. populations, suggesting evolution resistance these herbicides. Sanger sequencing target genes determined population carried a Trp-574-Leu mutation ALS gene an Arg-98-Gly PPX2 gene, while His-197-Pro (first documentation genus), but no was detected gene. Single-nucleotide polymorphism (SNP) genotyping developed enable efficient screening future order limit spread herbicide-resistant populations. In addition, preliminary testing revealed three mutations ubiquitous respective These two represent first cases United States, as well this particular herbicide-resistance profile inhabiting America. While found population, we suggest has evolved herbicides, mechanism is be determined.

Protein trans-splicing to produce herbicide-resistant acetolactate synthase.
Protein splicing in trans has been demonstrated both in vivo and in vitro by biochemical and immunological analyses, but in vivo production of a functional protein by trans-splicing has not been reported previously. In this study, we used the DnaE intein from Synechocystis sp. strain PCC6803, which presumably reconstitutes functional DnaE protein by trans-splicing in vivo, to produce functional...

ilvB-encoded acetolactate synthase is resistant to the herbicide sulfometuron methyl.
The herbicide sulfometuron methyl is a potent inhibitor of the branched-chain amino acid biosynthetic enzyme acetolactate synthase (ALS) isolated from bacteria, fungi, and plants. However, it did not prevent growth of wild-type Salmonella typhimurium LT2 or Escherichia coli K-12. These species each contain two acetolactate synthase isozymes. Growth of S. typhimurium and E. coli mutants lacking ...
متن کاملHerbicide-Resistant Mutations in Acetolactate Synthase Can Reduce Feedback Inhibition and Lead to Accumulation of Branched-Chain Amino Acids
The branched-chain amino acids (BCAAs) valine, leucine and isoleucine are essential amino acids that are critical for animal growth and development. Animals need to obtain BCAAs from their diet because they cannot synthesize them. Plants are the ultimate source of these amino acids. Acetolactate synthase (ALS) is the first common enzyme in the biosynthesis of BCAAs. The metabolic control of BCA...
متن کاملHerbicide resistances in Amaranthus tuberculatus: a call for new options.
Amaranthus tuberculatus is a major weed of crop fields in the midwestern United States. Making this weed particularly problematic to manage is its demonstrated ability to evolve resistance to herbicides. Herbicides to which A. tuberculatus has evolved resistance are photosystem II inhibitors, acetolactate synthase inhibitors, protoporphyrinogen oxidase inhibitors, and glyphosate. Many populatio...
